Girlfriend, wife, lover and ex-girlfriend are all terms that can be applied to a female love interest.

Girlfriend Your girlfriend is the girl who you're dating, almost always exclusively. She is the romantic interest in your life, but you are not engaged or married yet.

In most cases (other than where arranged marriage is concerned) a girl is your girlfriend before she becomes your wife.
If you have a girlfriend, then you are either a boyfriend or a girlfriend, depending on your gender.

Wife Your wife is the woman that you're legally married to.

Having a wife makes you a husband (or possibly a wife, in countries where gay marriage is legal).

Most people marry with the intention of staying with their partner for the rest of their life, although divorce is common. Married couples almost always live together.

Lover This one is a lot harder to define, as different people associate it with different things.

For most people, a lover is somebody that you are sexually intimate with and are in love with. However, somebody does not have to be your girlfriend or boyfriend to be your lover.

Lover is a gender-neutral term, unlike girlfriend, wife and ex-girlfriend, all of which are used to refer to females.

Ex-girlfriend An ex-girlfriend is a girl who used to be your girlfriend but is not your girlfriend anymore.

Most of the time, people will avoid their ex-girlfriends or ex-boyfriends, as it can be difficult to maintain a friendly relationship with somebody after breaking up with them.

A lover is male or female and is the person you love (romantically).
An ex-girlfriend is a girl who you use to date, but broke up with.
A fiance is the person (male or female) who you are engaged to (I know you did not ask about that, but I thought it might be helpful).
