Well he can ask her after all she is his wife and the main thing in marriage should be honesty and trust and if he is not sure he can have her go to a doctor and be checked up but that can lead to trust issues too. So just ask her first and if you can't find yourself believing her then ask her to take the test if she is she will if she isn't then she won't.
