I don't know, I think it might be a bit more complex than that...
- How much contact do you have with your crush in the first place?
- Is he/she likely to follow you back?
If you barely speak to someone at school but all of a sudden follow them on Twitter (especially if you're not following any other people you don't talk to) this could be seen as a sign of interest.
Yes, you want to make your interest clear eventually, but do you really want to do it over Twitter?
What if your crush doesn't follow you back?
Are you opening yourself up to public rejection if you don't get a follow back? What if you start tweeting her and she doesn't reply? Remember Twitter is a public platform, and you won't be able to DM until you get a follow back... Maybe Facebook would be a better platform in this instance.
So, overall, I'm not saying don't go for the follow... Just proceed with caution.
